摘要
本文以乌鱼(Ophrocepholus argus)肝为材料提纯了线粒体DNA.用四种限制性核酸内切酶:EcoR Ⅰ,BamH Ⅰ,Pst Ⅰ和Bgl Ⅱ分别进行完全酶解,各产生1、2、2、和2个片段.用六对限制酶:EcoR Ⅰ/BamH Ⅰ,EcoR Ⅰ/pst Ⅰ,EcoR Ⅰ/Bgl Ⅱ,BamH Ⅰ/Bgl Ⅱ,BamH Ⅰ/Pst Ⅰ和Bgl Ⅱ/Pst Ⅰ完全酶解分别得到3,3,3,3,4和4个片段.测得乌鱼肝线粒体DNA分子量为9.99×10~6道尔顿,长16.2kb.组建了四种限制酶图谱,以pBR_(325)质粒为载体,E colli HB101为受体菌,克隆了乌鱼肝线粒体DNA 1.8kb EcoRⅠ/BamH Ⅰ片段.
Snakeheaded fish liver mtDNA was digested completely by four different restiction endonuclea- ses. The cleavage sites are as follow: l for EcoR 1, 2 for BamH 1, 2 for Pst I and 2 for Bgl 1 Afolec- ular weight of the mtDNA was found to be 9.99×10~6 daltons about 16.2 kb. The restriction endonu- clease map was constructed. With pBR325 as vector, the mtDNA 1.8 kb EcoR I/BamH I fragment was cloned.
