摘要
槐果碱(Sophocarpine,Sop)系从豆科槐属植物苦豆子(Sophora alopecuroides L.)中提取出来的生物碱,属于四环的喹诺里西啶类(Quinolizidine)生物碱。动物实验和临床应用表明,Sop具有平喘、抗肿瘤及抗心律失常等作用,该药毒性低,副作用小。本文采用高效液相色谱法(HPLC)测定家兔血浆中Sop的浓度,方法灵敏,结果准确,重现性较好,该方法已用于药代动力学的研究。
A rapid sensitive and specific method is proposed for the determination of sophocarpine in rabbit plasma by using a revefsed-phase HPLC column (0.26×25 cm, packed with ODS-HC SlL-X-I), a pre-column (0.26×7cm, packed with ODS-HC SIL-X-II) and UV detector at 215nm. The mobile phase was methanol-water (100:15. containing lμ1 triethylamine per 100ml)land the flow rate was 1.0 ml/min. Cinchonidine was used as an internal standard, and peak heights were used for quantitation. Sophocarpine was extracted from rabbit plasma by liquid-liquid extraction. The recovery of sophocarpiae from plasma was 99.34%. The minimum detectable concentration was 0.08 μg/ml ia plasma, and the CV values were all less than 5%.
