氯苯氨丁酸对慢性应激大鼠心血管功能的影响 被引量:1

摘要 目的:观察氯苯氨丁酸对应激大鼠心血管功能的影响,进一步探究其抗应激作用。方法:实验选用成年健康大鼠50只,随机将50只大鼠分为对照组,应激1,6,12和15d组,每组10只。对照组大鼠单独饲养,避免外界干扰;应激组大鼠采用足底电击结合噪声制备应激性高血压大鼠模型,所有实验大鼠经腹腔注射γ-氨基丁酸B型受体激动剂氯苯氨丁酸(4~6mg/kg),用PowerLab生理记录仪观察动物的心血管功能变化。结果:对照组、应激1,6,12和15d组的大鼠在给药前的动脉压分别为(120±17),(128±9),(141±16),(130±10),(132±13)mmHg,注射4mg/kg氯苯氨丁酸对血压和心率无明显影响;注射6mg/kg氯苯氨丁酸时各组动物的动脉压分别降低到(70±13),(82±18),(86±11),(79±10),(78±12)mmHg,与给药前比较,差异有显著性意义(t=10.45~12.43,P<0.01),对心率无明显影响。结论:氯苯氨丁酸能够降低正常和应激大鼠血压,有剂量依赖性,氯苯氨丁酸对应激性高血压具有降压作用。 AIM:To observe the influence of baclofen on the cardiovascular function of chronic stressing rats, so as to further study its antistress effect. METHODS:Fifty healthy adult rats were randomly divided into control group,1,6,12 and 15-day stress groups,there were 10 rats in each group.Rats in the control group were fed separately in avoidance of outside interference,while those in the stress groups were made into models of stress hypertension by electronic shock of foot and noise.All the experimental rats received intraperitoneal injection of gamma-aminobutyric acid B receptor(GABAB receptor) agonist baclofen(4 to 6 mg/kg),and the changes of cardiovascular function were detected with the PowerLab physiological instrument. RESULTS:The arterial pressure of rats in the control group,1,6,12 and 15-day stress groups before medication were(120±17),(128±9),(141±16),(130±10),(132±13) mm Hg respectively;Injection of 4 mg/kg baclofen had no obvious influence on blood pressure and heart rate;As compared with before medication,the arterial pressure significantly reduced to(70±13),(82±18),(86±11),(79±10),(78±12) mm Hg after injection of 6 mg/kg baclofen(t=10.45 to 12.43,P< 0.01),while it had no obvious effect on heart rate. CONCLUSION:Baclofen can decrease the blood pressure of normal and stress-induced rats,and it is dose-dependent.Baclofen had the effect of decreasing blood pressure depress in rats with stress-induced hypertension.
