摘要
目的:观察异丙肾上腺素(isoproterenol,ISO)致大鼠急性心肌损伤早期免疫分子E-选择素和单核细胞趋化因子1(monocytechemoattractantprotein-1,MCP-1)基因表达,研究N-乙酰半胱氨酸(N-acetyl-L-cysteine,NAC)对急性心肌损伤的保护作用及作用机制。方法:实验选用雄性Wistar大鼠30只,随机将30只大鼠分为对照组、ISO组和ISO+NAC组,每组10只。测定大鼠血清肌酸激酶、乳酸脱氢酶(lactatedehydrogenase,LDH)含量,测定心肌组织抗氧化体系中超氧化物歧化酶(superoxidedismutase,SOD)和谷胱甘肽-过氧化物酶(glutathioneperoxidase,GSH-Px)的活性;测定心肌组织中E-选择素、MCP-1mRNA的表达。结果:ISO组大鼠心肌组织SOD和GSH-Px的活性显著低于对照组(P<0.01);ISO组大鼠心肌组织中E-选择素、MCP-1mRNA表达显著高于对照组(P<0.01);ISO+NAC组心肌组织的SOD和GSH-Px活性犤(3.06±0.44)mkat/g,(65.85±9.84)μkat/g犦显著高于ISO组犤(1.74±0.36)mkat/g,(35.51±5.67)μkat/g犦(P<0.01);而心肌组织中E-选择素、MCP-1mRNA表达显著低于ISO组(P<0.01)。结论:NAC对ISO致大鼠急性心肌损伤有保护作用,其机制可能与增加抗氧化酶的活性,减少心肌组织中E-选择素、MCP-1基因的表达有关。
AIM:To observe the change of immune factors E-selection and monocyte chemoattractant protein-1(MCP-1) gene expression induced by isoproterenol during early acute heart injury in rat,and study the protective effects of N-acetyl-L-cysteine(NAC) on acute heart injury and its mechanisms. METHODS:Thirty male Wistar rats were randomly divided into three groups with 10 in each: isoproterenol (ISO) group,ISO+NAC group and control group.Serum contents of creatine kinase(CK) and lactate dehydrogenase(LDH),activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in the antioxidative system of myocardial tissue,and myocardial E-selection and MCP-1 mRNA expression were examined. RESULTS:Compared with control group,ISO group had significantly lower myocardial activities of SOD and GSH-Px,but had significantly higher myocardial E-selection and MCP-1 gene expression.The myocardial activities of SOD and GSH-Px in the ISO+NAC group[(3.06±0.44) mkat/g,(65.85±9.84) μkat/g] were remarkably higher than those in the ISO group[(1.74±0.36) mkat/g,(35.51±5.67) μkat/g](P< 0.01),while myocardial E-selection and MCP-1 gene expression were significantly lower in the ISO+NAC group than in the ISO group(P< 0.01). CONCLUSION:NAC has the protective effects on acute heart injury induced by ISO,which is probably associated with the improving activity of antioxidant enzymes,decreasing E-selection and MCP-1 gene expression.
