枸杞子对人成纤维细胞寿命及c-fos基因表达的影响 被引量:14

摘要 目的:检测枸杞子提取液(fructuslyciiextract,FLE)对人二倍体成纤维细胞国内标准株(2BS)寿命及c-fos基因表达的影响。方法:实验分两组,25代龄2BS细胞用单纯DMEM培养基培养为对照组,实验组25代龄2BS细胞用0.025%FLE培养,计数实验组及对照组所培养的代龄。制备c-fos探针,并以Northern杂交的方法检测c-fos在0.025%FLE处理后的表达情况。结果:实验组的代龄(61.0±2.9)代明显大于对照组(49.0±2.5)代(t=3.31,P<0.05)。在衰老细胞中发现经0.025%FLE处理后能检测到c-fos的表达,而对照组未检测到。结论:枸杞子能有限的延长2BS细胞寿命,并对衰老2BS细胞c-fos基因转录有诱导作用,为延缓细胞衰老的基因学研究提供理论基础。 AIM:To detect the influence of fructus lycii extract(FLE) on the lifespan of the human fibroblasts diploid cell 2BS strain and the expression of c-fos gene. METHODS:The 25-generation 2BS cells were incubated with simple DMEM culture medium(control group) and 0.025%FLE(experimental group) respectively,and the cultured generations were counted in the experimental group and control group.The c-fos probe was prepared, and the expression of c-fos gene after treatment of 0.025%FLE was detected with the northern blotting method. RESULTS:The lifespan was obviously greater in the experimental group[(61.0±2.9) generations] than in the control group[(49.0±2.5) generations](t=3.31, P< 0.05).After treated with 0.025%FLE,the expression of c-fos could be detected in the aging cells,while could be observed in the control group. CONCLUSION:FLE can limitedly prolong the lifespan of the 2BS cells,and can induce the transcription of c-fos gene in aging 2BS cells,and provides theoretical basis for the genetic study of postponing cellular aging.
