Smad在牙本质涎磷蛋白基因表达中的作用

摘要 目的:研究Smad蛋白在牙本质涎磷蛋白(dentinsialophosphoprotein,DSPP)基因表达中的作用。方法:细 胞培养,瞬时转染和报告基因检测。结果:转化生长因子β1(transforminggrowthfactor-β1,TGF-β1)抑制DSPP 基因启动子活性。过表达野生型Smad3蛋白显著增强了TGF-β1对DSPP基因表达的转录抑制,而过表达Smad3 突变型蛋白则显著抑制了TGF-β1对DSPP基因启动子活性的影响。过表达Smad2野生型或突变型蛋白对TGF -β1抑制DSPP基因启动子活性无明显影响。结论:Smad3在介导TGF-b1对DSPP基因的转录调控中发挥重要 的作用。 Objective: To characterize the role of Smad proteins in transcriptional regulation of dentin sialophosphoprotein (DSPP). Methods: Cell culture, transient transfection and luciferase assay. Results: Transforming growth factor-β1 (TGF-β1) inhibited the activity of DSPP promoter luciferase reporter construct containing the sequence between -791 bp and +54 bp of mouse DSPP gene. Overexpression of wild-type Smad3 potentiated the inhibitory effect of TGF-β1 on transcriptional regulation of DSPP, while dominant negative Smad3 decreased the effect. In contrast to Smad3, wild-type Smad2 or its dominant negative mutant had little effect on TGF-β1 regulation on the promoter activity of DSPP. Conclusions: The results indicated that the crucial role of Smad3 in mediating transcriptional regulation of DSPP by TGF-β1.
