摘要
目的 :检测P5 3、MDM 2、P2 1WAFI、BAX在食管鳞癌中的表达 ,探讨它们与食管鳞癌临床病理特征之间的关系。方法 :采用免疫组化检测食管鳞癌、正常食管粘膜和不典型增生组织中上述基因蛋白的表达。结果 :各指标在上述组织中的表达有显著性差异。MDM2在淋巴结转移组中的阳性表达率为 80 %,未转移组中为 35 .7%,两者之间有显著性差异。随着食管鳞癌组织学分级的增高 ,P5 3、MDM2的表达逐渐增高 ,P2 1WAF1、BAX的表达逐渐降低。结论 :P5 3、P2 1、MDM2、BAX多个基因的异常在食管癌的发生、发展过程中共同发挥作用 ;MDM2过表达与肿瘤的侵袭性和转移有关。P5 3、P2 1WAF1、MDM 2在食管鳞癌中的表达无相关性 ,P5 3与BAX的表达呈负相关。
Objective: To determine the expression of P53, P21WAFI, MDM2, and BAX in esophageal squamous cell carcinoma(ESCC). Methods:The expressions of these proteins were examined by immunohistochemical method in esophageal squamous cell carcinomas, normal and dysplastic esophageal mucosa.Results:The expression of every protein had significant difference among esophageal squamous cell carcinoma, dysplastic and normal esophageal mucosa, of which P53 and MDM2 enhanced gradually but P21 WAFI and BAX decreased gradually. The positive rate of MDM2 in lymph node metastasis was 80% and in non-lymph node metastasis was 35.7%. The expression of P53 and MDM2 increased but P21 WAF1 reduced with the stage rising in squamous cell carcinoma. Conclusion:Abnormality of P53,MDM2,P21 and BAX may be involved in occurrence and development of esophageal squamous cell carcinoma. Overexpression of MDM2 is related to infiltration and metastasis. The expression of P21 WAF1 and MDM2 may have non-P53 dependent pathway in esophageal squamous cell carcinoma. The expression of P53,P21 and MDM2 has no correlationship.
