摘要
目的 研究外源性白细胞介素 10 (IL - 10 )干预对实验性大鼠肝纤维化肝星形细胞表达α-平滑肌肌动蛋白 (α- SMA)和核因子 -κB(NF-κB)的影响。 方法 6 0只清洁级 SD大鼠 ,随机分为正常对照 (N组 )、肝纤维化 (C组 )和 IL - 10干预 (I组 )。以 CCl4 腹腔内注射构建肝纤维化模型 ,并予 IL - 10腹腔内注射干预造模。于第 7周和第 11周分别随机选取一批大鼠 ,分离肝星形细胞 (HSC) ,以免疫细胞化学法检测各组 HSC中α- SMA和 NF-κB表达情况。 结果 N组α- SMA和 NF-κB仅少量表达且着色浅 (阳性率分别为 36 .5 %和 2 8.5 % ) ,免疫化学评分分别为0 .6 6± 0 .0 7和 0 .4 2± 0 .0 5 ;C组所有细胞均表达α- SMA和 NF-κB且表达明显增强 ,第 7周二者评分分别为 2 .4 3± 0 .0 3和 1.73± 0 .0 9,第 11周为 2 .4 7± 0 .14和 2 .4 8± 0 .70 (与 N组比较 ,P<0 .0 1) ;I组二者表达较 C组明显减弱 ,第 7周时评分分别为 2 .14± 0 .11和 1.6 2± 0 .1(P<0 .0 5 ) ,第 11周为 2 .0 9± 0 .0 6和 1.92± 0 .4 2。随肝纤维化进展 ,C组中第 11周 NF-κB的表达较第 7周有增加趋势 (P<0 .0 1) ,而α- SMA的表达不随纤维化的进展而变化 (P>0 .0 5 )。 结论 外源性 IL -
Objective To study the effects of interleukin-10 on the expression of α-smooth muscle actin(α-SMA) and nuclear factor-κB(NF-κB) in hepatic stellate cells in experimental rat hepatic fibrosis. Methods 60 SD rats were divided into control group(group N), the model group(group C) and the IL-10 treated group(group I) randomly. Rat hepatic fibrosis model induced by CCl 4 was established, and treated with IL-10 from the 3rd week. Hepatic stellate cells were isolated from these rats at the 7th and 11th weeks during the course of liver fibrosis, respectively. The expression of α-SMA and NF-κB in HSC were measured by S-P immunohistochemistry. Results The positive expression of α-SMA and NF-κB in group N was 36.5% and 28.5% respectively and the positive immunochemical score was (0.64±0.05) and (0.43±0.08) respectively. The positive levels for α-SMA and NF-κB in group C were increased significantly compare to group N(P<0.01). Their positive score in group C in the 7th week was (2.43±0.03) and (1.73±0.09), respectively, in the 11th week the score was (2.47±0.14) and (2.48±0.70), respectively. The positive score decreased significantly(P<0.05) in group I. In the 7th week and the 11th week, their positive score was (2.14±0.11) and (1.62±0.10), (2.09±0.06) and (1.92±0.42), respectively. In the 11th week, the HSC in group Ⅰ became round and the nuclear pyknosis. Conclusion The positive expression of α-SMA and NF-κB in hepatic stellate cells was decreased by ectogenic IL-10 in liver fibrosis induced by CCl 4. The IL-10 inhibit the activation of hepatic stellate cells.
