摘要
目的 探讨cyclinD1和p2 7蛋白在血管瘤发生、发展及退化过程中的表达状况及其意义。方法 采用免疫组织化学方法 (S-P法 )检测人皮肤血管瘤增生期、退化期及正常皮肤组织中cyclinD1和 p2 7的表达水平 ,并结合第Ⅷ因子相关抗原的免疫组织化学染色证实表达cyclinD1和 p2 7的细胞是血管内皮细胞。利用计算机图像分析技术测量不同时期血管瘤组织和正常皮肤组织cyclinD1和p2 7表达的平均光密度和平均阳性面积率。 结果 增生期血管瘤内皮细胞cyclinD1表达水平高于退化期 ,差异有显著意义 (P <0 .0 5 ) ,退化期血管瘤内皮细胞cyclinD1表达水平与正常皮肤组织相比 ,差异无显著意义 (P >0 .0 5 ) ;而增生期血管瘤内皮细胞p2 7表达水平低于退化期 ,差异有显著意义(P <0 .0 0 5 ) ,退化期血管瘤内皮细胞 p2 7表达水平与正常皮肤组织相比 ,差异有极显著意义 (P <0 .0 0 1)。 结论 cyclinD1和p2 7可能通过介导血管瘤内皮细胞增殖和血管生成而在血管瘤的发生、发展及退化过程中起重要作用。
Objective To investigate the expression and significance of cyclin D1 and p27 in the occurrence, development and regression of human dermal hemangiomas. Methods The immunohistochemical technique was used to examine the expression of cyclin D1 and p27 in proliferating, involuting human hemagiomas and nomal skin. Immunohistochemical technique for factor Ⅷ-related antigen was used to prove that the cells which expressed cyclin D1 and p27 are endothelium. Image analysis system was applied to measure the expressing level of cyclin D1 and p27 in different stage of hemangiomas and in normal skin tissues. Results The expression of cyclin D1 is significantly higher in proliferating hemangiomas than in involuting hemangomas (P<0.05), but is not significantly higher in the involuting hemangiomas than in normal skin tissues(P>0.05); The expression of p27 is significantly lower in proliferating hemangiomas than in involuting hemangomas(P<0.005), and is also significantly lower in the involuting hemangiomas than in normal skin tissues(P<0.001). Conclusion It is suggested that cyclin D1 and p27 might play an important role in the proliferation of human hemangiomas endothelial cells and angiogenesis.
