摘要
[目的 ]研究丙烯腈 (AN)的雌激素样作用。 [方法 ]采用经典大鼠子官增重测试方法 ,对去卵巢SD大鼠皮下注射剂量分别为 0、10、15、2 5、40、60mg/kg受试物丙烯腈 ,每天 1次、连续 3d ,同时皮下注射剂量为 0 .0 0 1mg/kg雌二醇。同时设雌二醇为阳性对照组和去离子水为溶剂对照组。最后 1次给药 2 4h后 ,将大鼠麻醉后取血并处死 ,观察受试动物子宫湿重、干重及肝脏、肾脏重量等指标。 [结果 ]丙烯腈各剂量组大鼠子官干重、湿重与溶剂对照组差异无显著性(P >0 .0 5 ) ,阳性对照组与溶剂对照组及丙烯腈各剂量组比较 ,子宫干重、湿重差异有显著性 (P <0 .0 5 )。丙烯腈各剂量组除最高剂量组肝脏重量有轻微增加外 (P <0 .0 5 ) ,受试动物未出现明显系统毒性。 [结论
Objective To investigate the estrogenic effect of acrylonitrile(AN) by the classic uterotrophic assay in rats. Acrylonitrile was injected subcutaneously at the doses of 0,10,15,25,40 and 60 mg/kg,once a day for 3 days,to ovaeriectomized Sprague-Dawley(SD) rats.Ethynylestradiol(EE) was also injected subcutaneously at a dose of 0.001 mg/kg as a positive control.All rats were killed 24 h after the last administration.The wet or blotted uterine weights,kidney and liver weights of rats were recorded. There were essentially no differences in the wet or blotted uterine weights between the control and any of the AN-treated groups(P>0.05).The positive control ethynylestradiol(0.001 mg/kg per day) showed a significant effect using this assay protocol(P<0 05).There was no excessive systemic toxicity in this assay except for slightly increased liver weight in the highest AN-treated group(P<0 05). [Conclusion] Uterotrophy has not been observed in ovariectomized rats treated subcutaneously with AN up to 60 mg/kg.
