摘要
采用ALLTEC LiChrospher Si60色谱柱,以正己烷/异丙醇为流动相,通过叔丁基甲醚对维生素D2进行萃取,直接进行HPLC分离,对脂肪乳剂中维生素D2进行准确地测定。
A rapid and convenient HPLC method for the determination of vitamin D2 in fat emulsion was developed. Vitamin D2 in fat emulsion was extracted completely with tert-butylmethylether. After dehydration with anhydrous sodium sulfate and phase-separation by centrifugation at 3 000 r·min^(-1), 25 μL of the clear upper-layer solution was taken for HPLC determination. The Alltech LiChrospher Si 60 chromatographic column was used in the analysis and a mixture of n-hexane and iso-propanol was used as mobile phase. Result of determination was calculated by the peak heights with reference to the peak heights of the standard solutions, RSD and average recovery of this method were found to be 1.2% (n=6) and 100.3% (n=10) respectively. Results obtained in the analysis of 12 samples checked very well with those obtain by the standard method.
