高效液相色谱法测定蓝绿藻中微囊藻毒素 被引量:9

摘要 以铜绿微囊藻为研究对象,考察了不同溶剂、不同超声波震荡时间和不同固相萃取(SPE)洗脱液浓度对藻毒素提取效率的影响,以及梯度淋洗HPLC分离藻毒素条件的优化。藻毒素浓度在1~100 mg·L-1范围内,峰面积与藻毒素进样量呈线性关系,相关系数为0.999 5,检出限为0.12 ng。方法可靠、灵敏度较高,用于实际藻样中藻毒素的测定,回收率为96.5%~98.0%。 Effect of toxicant, microcystin, different solvents, ultrasonic treatment time and eluants on the extraction efficiency of microcystis aeruginosa was studied. Conditions of gradient elution and HPLC separation of the toxicant were also optimized. It was found that linear relationship between peak area and concentration of microcystin holds in the range of 1~100 mg·L^(-1), having the regression equation as follows: y=1 478 x+(3 884.3) (r=0.999 5). Detection limit (S/N=3) of the method was found to be 0.12 ng. RSD′s (n=6) was 3 8%, and average recovery obtained was 98.0%.
