摘要
利用指示植物巴西牵牛接种传毒试验及电镜负染检测,从我省表现斑驳花叶与明脉症状的甘薯病毒样品中,首次分离出甘薯羽状斑驳病毒。笔者采用5种病毒抗体(IgG)、两种血清学方法(DAS-ELISA和NCM-ELISA),对四川成都、绵阳、南充、内江等地的158份甘薯病毒样品进行了检测,结果表明,四川以上地区的甘薯普遍存在甘薯羽状斑驳病毒(SPFMV)和甘薯潜隐病毒(SPLV)。应用脱毒苗露地栽培和小区产量对比试验证明,脱毒甘薯增产效果较稳定;笔者对甘薯茎尖脱毒及组织培养方法进行了探索。
Using grafting inoculation with indicator or plat Ifomoea setosa and leaf-dip method of electron micoscopic observation, Sweet Potato Feather Mottle Virus (SPFMV)was for first time isolated and identified from virus samples with mosaic and veinbinding symptoms in Sichuan province. 158 virus samples collected from Chengdu, Mianyang, Nanchong and Neijiang of Sichuan province were assayed by DAS-ELISA and NCM-ELISA of 5 sweet potato virus antibodies (IgG) . The serological detection results indicated that the SPFMV and Sweet Potato Latent Virus (SPLV) exist far and wide in the above areas in Sichuan. The results of the test of plot-yield between virus-free sweet potatos and check proved that the increasing effect of virus-free potato planted in open fields and nurseries was much stable for two years. The authors have also done some research in the method of virus-free meinstem culture and culture conditions.
