摘要
目的 初步探讨 pH、Tris-HCl浓度、十二烷基硫酸钠 (SDS)与 β-环糊精 (β -CD)浓度的比值三者变化对重组内抑素复性的影响。方法 根据人工分子伴侣对重组蛋白质的复性机理 ,采用人工分子伴侣方法应用于重组内抑素的复性。结果 通过优化后确定了其最佳复性时条件为 :pH为 8 0 ,Tris -HCl浓度为 10 0mM ,SDS∶β -CD为 1∶4 ,复性回收率可达89 %。
Objective To investigate the recombinant endostatin refolding from the effects of Tris-HCl concentration,pH,and the ratio of sodium dodecyl sulfate(SDS) to beta-cyclodextrin(β-CD).Methods Based on the reactive principle of molecular chaperone,we adopted an artificial molecular chaperone-assisted refolding method for recombinant endostatin refolding.Results The results proved that the optimal refolding condition was as follows:pH was 8.0,Tris-HCl concentration was 100 mM and the concentration ratio of SDS to β-CD was 1∶4.The recycle rate was 89%.Conclusion This method is suitable for recombinant endostatin refolding under optimized condition.
