摘要 本论文对气相色谱法、地衣酚-盐酸法、间苯三酚-冰醋酸法以及Duffau蒸馏法四种不同的戊聚糖含量测定方法进行研究。在研究过程中以气相色谱法为标准,就地衣酚-盐酸法、间苯三酚-冰醋酸法以及Duffau蒸馏法与气相色谱法之间的相关性进行研究,通过研究得到测定谷物中戊聚糖含量的最佳方法,即采用间苯三酚-冰醋酸比色法。研究发现间苯三酚-冰醋酸法与气相色谱法具有较好的相关性,并且该方法测定较快速、准确。并对苯三酚-冰醋酸法测定戊聚糖含量的测定条件进行优化,得到该方法测定的最适条件为:测定样品与抽提试剂的共沸反应时间为30min;采用单波长测定,测定波长为552mm;另外样品中己糖含量对测定结果影响不大。 Four different determination methods for cereal pentosans were studied compared atively in this paper. The four methods were gas chromatography method (GL method ) , Orcind - HC1 method, phtoroglucind - acetil acid (Douglas'method)and duffan distillation method. The correlation between different methods were analyzed using gas chromatography method as determining standard. The comparative study showed that Douglas' method was preferred for determination of pentosans for its simplicity, high - speed, and accaracy. The condition to the determination were also optimized. The optimized resalts indicateol that in the determining process of pentosan using Donglas'meth-od,the reaction time between sample and extracting solution at 100℃ waw 30min. The determination ware length was 552nm. In addition, hexose has little effect to pentosan content of determined sample.
