摘要
以Brunt法提取的水仙花叶病毒(Natcissus Mosaic Virus,NMV)为抗原,免疫家兔获得抗血清,经35%饱和度的(NH_4)_2SO_4提纯后与辣根过氧化物酶共价交联,该免疫酶标抗体能与水仙叶片表皮细胞、鳞茎片表皮细胞内的病毒呈特异结合,并催化底物DAB和H_2O_2产生棕色沉淀反应而别于对照组;可作为水仙植株存在NMV的快速、简便的检测手段。
Immunity rabbit acquires antiserum for it obtains Narcissus Mosaic Virus exiracted by Brunt method as its antigen. After purifying by 35% saturatrd (NH4)2SO4, the antiserum crosslinks covalet with horseradish peroxidase. This immunoenzyme antibody exceptionally conjugate with virus in leaf blade epidermal cells and balb blade epidermal cells of Narcissus tazatta, then catalyzied substrate DAB and H2O2. Thus brown precipitation reaction appears and it is different from the control groups. It can act as a rapid, convenient detective method if the Narcissus tazetta plant exists Narcissus Mosaic Virus.
