摘要
目的 研究亚低温对小鼠局灶性脑缺血急性期的保护作用。方法 2 4只小鼠随机分为亚低温组和常温对照组 ,采用线栓法制作可逆性小鼠大脑中动脉缺血 (middlecerebralarteryocclusion ,MCAO)模型 ,缺血 30min 再灌注 2 4h ,亚低温组在MCAO后 1h开始给予 34.5℃ - 35℃亚低温处理 2h。分析两组小鼠脑组织含水量以及神经功能缺陷评分的变化。结果 亚低温组脑组织含水量在亚低温后 3h、2 4h的较常温对照组显著降低 (P <0 .0 5 ) ;亚低温组在亚低温后 3h、6h、12h、2 4h神经功能缺陷评分较对照组显著降低 (P <0 .0 5 ,P <0 .0 1) ;脑组织含水量与相应的神经功能缺陷评分具显著相关性 (P <0 .0 5 )。结论 亚低温对小鼠急性期局灶性脑缺血具有降低脑水肿。
Objective To investigate the effects of mild hypothermia on acute focal cerebral ischemia in mouse.Methods The model of reversible middle cerebral artery occulusion (MCAO), which had been ischemic for 30 min and reperfused for 24 hours, was made in 24 mice. Twenty-four mice were randomized into two groups: 12 mice were treated with the core temperature reduced to and maintained at 33°C to 35°C for 2 hours by surface cooling in 1 hour after MCAO occurred as the mild hypothermia group, and the other 12 mice were treated without hypothermia as the control group. Brain water content and neurological deficit scores were evaluated. Results Results The brain water content in the hypothermia group was significantly lower than that in the control group at 3 and 24 hours after hypothermia treatment (%P%<0.05). The neurological function scores of the hypothermia group were very lower than those of the control group at 3, 6, 12 and 24 hours after hypothermia treatment (%P%<0.05).Conclusion Mild hypothermia can reduce the cerebral edema and attenuate the neurological deficits in mice with acute focal cerebral ischemia.
