脂联素基因多态性与2型糖尿病的关系 被引量:27

Relationship between adiponectin gene polymorphisms and type 2 diabetes

摘要 目的研究脂联素基因单核苷酸多态性 (SNP) 4 5T→G和 2 76G→T两个位点与上海地区汉族人 2型糖尿病之间的关系。方法采用病例—对照研究方法 ,以聚合酶链式反应—限制性内切酶长度多态性 (PCR RFLP)技术 ,对 195例 2型糖尿病患者和 187例正常对照者脂联素基因SNP4 5、SNP2 76多态性位点进行基因分型。结果SNP4 5和SNP2 6 7两个多态性位点的基因型和等位基因频率在 2型糖尿病组和正常对照组中的分布无显著差异 (P >0 .0 5 ) ;各组中TG单倍型纯合携带者 (TG/TG)与TG单倍型杂合携带者 (TG/X)或未携带者 (X/X)的体重指数比较均无显著性差异 (P >0 .0 5 )。结论脂联素基因的SNP4 5和SNP2 76多态性位点与上海地区汉族人群中 2型糖尿病无明显相关性。 Objective To test the association of single nucleotide polymorphisms (SNP)45T→G and SNP276G→T in the adiponectin gene with type 2 diabetes in Shanghai Han population. Methods For the case-control study, PCR-RFLP method was used to determine the distributions of allele and genotype frequencies of SNP45T→G polymorphism in exon 2 and SNP276G→T polymorphism in intron 2 in adiponectin gene in 195 type 2 diabetic patients and 187 normal control subjects. Results The distribution of genotype and allele frequencies of the SNP45 and SNP276 polymorphism were not different in diabetes and control groups(P>0.05). There also was no difference in BMI between homozygous carries of the TG haplotype and heterozygous carries of the TG haplotype(TG/X) or noncarries(X/X) in diabetes and control groups(P>0.05). Conclusion The results suggest that these two SNPs in adiponectin gene were not associated with type 2 diabetes in Shanghai Han population.
