粒细胞集落刺激因子修复心肌梗死区域的实验研究 被引量:1

Effects of granulocyte colony-stimulating factor on the regeneration of myocardial infarction in rabbits

摘要 目的 研究粒细胞集落刺激因子对新西兰白兔心肌梗死区域的修复作用。方法  30只新西兰白兔随机分为假手术组、心肌梗死组及药物治疗组。药物治疗组心肌梗死模型制作采用开胸左冠脉边缘支结扎法 ,术前 2 4h及术后 6d予G CSF 10 μg/ (kg·d)皮下注射处理。另外两组皮下注射等量的生理盐水 1周对照。三组术前、术后 4周均查心超 ,观察心功能的改变。 4周时处死动物 ,取出心脏 ,HE和免疫组化染色观察心肌梗死区域的病理变化。结果 药物治疗组 4周后心功能EF值 (0 6 95± 0 0 38)较心肌梗死组(0 5 5 4± 0 0 6 5 )上升 (P <0 0 5 ) ,低于假手术组 (0 74 7± 0 0 19) ;HE染色和免疫组化病理检查发现有新生的血管。结论 粒细胞集落刺激因子可修复心肌梗死区域 ,促进血管新生 ,改善心功能。 Objective To investigate the effects of granulocyte colony-stimulating factor (G-CSF) on the regeneration of myocardial infarction in experimental rabbits.Methods Thirty New Zealand rabbits were randomly divided into sham-operation (SO) group, myocardial infarction(MI)group and G-CSF treatment (GT) group. Myocardial infarction was induced by ligation of coronary artery. The rabbits of GT group were treated with G-CSF 10 μg/kg subcutaneously before 24 h of ligation of coronary artery and for 6 days after surgery once a day. Those of SO group and MI group were received saline. Ecocardiography was performed in each group before surgery and after 4 weeks. The hearts were harvested for HE and immunohistochemistry pathological examination. Results EF of GT group (0.695±0.038) were higher than MI group(0.554±0.065 ) and lower than SO group(0.747±0.019 ) after 4 weeks(P<0.05). New vascular structures were also found in the infarct zones .Conclusion G-CSF treatment can be beneficial to the regeneration of vascular structures and improve cardiac function.
