β-1,4-半乳糖基转移酶V对胶质细胞瘤细胞SHG-44粘附的影响

摘要 目的 :研究 β 1,4 半乳糖基转移酶V(β 1,4 GalTV)对星形胶质瘤细胞株粘附能力的影响及其与细胞恶性行为的关系。方法 :将转染正义、反义 β 1,4 GalTV和空载体的SHG 4 4细胞株接种于包被有纤连蛋白 (FN)或层连蛋白 (LN)的板上 ,观察细胞粘附力的变化。采用WesternBlot的方法检测整合蛋白 β1表达变化及局部粘着斑激酶 (FAK)的蛋白水平和FAK的酪氨酸磷酸化水平。结果 :在FN和LN包被的板上 ,与转空载体的SHG 4 4细胞株相比 ,转正义 β 1,4 GalTV比转反义 β 1,4 GalTV的细胞在纤连蛋白和层连蛋白上的粘附能力增高 ,β1蛋白表达水平升高 ,FAK及其酪氨酸磷酸化水平增强。结论 :β 1,4 GalTV对SHG 4 4细胞的粘附能力确有影响 ,提示 β 1,4 Objective:In order to study whether β-1,4-galactosyltransferase V(β-1,4-GalT V) is related to the malignant behavior of SHG- 44 cells,the effects of β-1,4-GalT V on cellular adhesion of SHG- 44 cells was investigated in vitro.Methods:Cell adhesion assay was performed in this study.SHG- 44 cells transfected with sense,antisense β-1,4-GalT V and vector were cultured on the culture plates coated with fibronectin or laminin,adhesion ability was investigated,and integrin β1,focal adhesion kinase(FAK),and FAK tyrosine phosphorylation were detected by Western Blot.Results:Compared to SHG- 44 cells transfected with vector,the cellular adhesion ability and the expression level of integrin β1,FAK and FAK tyrosine phosphorylation of SHG- 44 cells which were transfected with sense β-1,4-GalT V cultured on fibronectin or laminin coated plates increased,and those of SHG- 44 cells transfected with antisense β-1,4-GalT V decreased.Conclusion:β-1,4-GalT V affects the adhesion ability of SHG- 44 cells, which suggests that β-1,4-GalT V might be involved in the malignant potential of tumor.
