摘要
目的 观察骨髓间充质干细胞 (mesenchymalstemcells ,MSCs)经 5 氮杂胞苷 (5 Aza)诱导分化为成骨骼肌肌细胞的过程。方法 分离纯化 4~ 6周龄SD大鼠的骨髓MSCs ,以 5 Aza作用 2 4h。于作用 1周后的不同时相点 ,观察细胞形态变化 ,免疫组织化学染色观察细胞内结蛋白 (desmin)、骨骼肌肌动蛋白 (actin)、肌球蛋白重链 (MHC)、肌生成素 (myogenin)表达。结果 经 5 Aza诱导后 14d ,部分细胞表达desmin ;诱导后 2 4d ,细胞表达myogenin、MHC、actin。细胞形态和排列方式发生明显变化 ,可见有肌管样细胞出现。结论 MSCs经 5 Aza诱导后可以向成骨骼肌细胞定向分化。
Objective To observe skeletal muscle cells derived from bone marrow mesenchymal stem cells (MSCs) induced by 5-azacytidine. Methods MSCs of 4-6 weeks old rats were separated and purified, and then were administered by 5-azacytidine for 24 hours. The morphological change, antigen expression of desmin, actin, MHC and myogenin evaluated by immunohistochemisty were observed at various intervals from 1 week on.Results MSCs began to express desmin at 14 days and myogenin, MHC,actin at 24 days after induction by 5-azacytidine. The morphology and alignment of the cells changed obviously. The myotube-like cells could be seen. Conclusion Myoblasts (skeletal muscle) could be differentiated from MSCs under the induction of 5-azacytidine.
