高效液相色谱测定黄瓜瓜条中赤霉素和脱落酸含量 被引量:32

摘要 本试验以秋露地栽培鲁黄瓜9号为试材,研究如何提取、纯化和利用高效液相色谱(HPLC)分析测定商品成熟期瓜条中赤霉素(GA3)和脱落酸(ABA)的含量。试材用液氮研磨以100%冷乙腈浸提,浸提液抽滤,氯仿去除色素,PVP去除酚类杂质,乙酸乙酯提取激素,最后过PT-C18预处理柱。以甲醇—水(含5%冰乙酸)为流动相,在μ-BandpakC18柱上进行分离,紫外检测器在252nm波长下检测、鉴定,并测得回收率分别为GA3:80 12%;ABA:81 33%。 Luhuanggua 9 was used as materials for detection of gibberellin abbeymycin (GA_3) and abscisic acid(ABA) by high-performance liquid chromatography(HPLC).The test materials were grinded in liquid nitrogen and lixiviated in acetonitrile,then pigment was eliminated in chloroform,phenolic was compounded in polyving pyrrolidone(PVP). Phytohormones were extracted in ethyl acetate, and finally PT-C_(18) column were used for further purification. GA_3 and ABA were separated and quantified by μ-Band pak C_(18) reversed HPLC with methanol-water at a flow rate of 0.8 ml/min.Detection was made by a UV detector at 252 nm.The frequency of reclamation was GA_3: 80.12% and ABA:81.33%.
