载荷对老年关节软骨组织凋亡基因Caspase-3 mRNA表达的影响

摘要 目的 对体外培养的老年关节软骨组织块进行不同性质压应力载荷干预,观察不同的载荷条件对关节软骨细胞凋亡过程的调控。方法 在体外对加工成一定几何形状、未纤维化的老年关节软骨组织块予以不同频率(0. 01、0 .1、1Hz与静止力)、载荷量(1. 5、1 .0与 0 .5MPa)的压应力,并通过反转录-聚合酶链反应法来观察不同应力条件下凋亡基因Caspase- 3mRNA的表达情况。结果 各应力组均有Caspase- 3mRNA的表达,其中对照组表达水平较低,中频各组表达与对照组无明显统计学差异;在静止应力组,以及高、低频组则有较强的表达。就表达强度而言,各载荷值无组间差异。结论 老年人关节软骨细胞的凋亡对于外界应力敏感,中频应力组对细胞的凋亡表现为抑制作用,其余各组则起到促进作用。 Objective To investigate the modulatory mechanism of the loads to apoptosis process of chondrocytes by interfering the senior articular cartilage tissue with several ranks of loads. Methods Using a set of special loading device, a series of senior, unfibrotic articular cartilage tissue explants was treated in vitro, which had been processed to a regular size and the provider had no bone or skeletal disease, loads with different frequency (0.01, 0.1,1 Hz and static load),and different magnitude (1.5,1.0 and 0.5 MPa). The mRNA expression odds of Caspase-3 were detected by the way of RT-PCR under different loads. Results There expressed Caspase-3 mRNA in each group designed, with low density of expression in the control, which was similar to the mid-frequency groups. The expression of the caspase-3 mRNA in the groups of high-frequency and low-frequency, included the static load group, all represented promotion result of the gene. But for the intensity of expression, the inter-group bias was inexistent. Conclusions The apoptosis of the senior articular chondrocytes is sensitive to the load suffered. Within all processed loads, the mid-frequency ones can yield the protective effect, while the others resulted in promotion.
