肝癌细胞系SMMC7721蛋白质组学初步分析 被引量:10

摘要 目的 分析肝癌细胞系SMMC772 1的蛋白质表达 ,筛选肝细胞癌的差异蛋白。方法 应用表面增强激光解吸离子化蛋白质芯片技术检测肝细胞癌细胞系SMMC772 1及正常肝细胞L0 2的蛋白质谱。用PBSII C型蛋白质芯片阅读机读取数据并进行分析。结果 与L0 2细胞相比 ,有 15个蛋白质在SMMC772 1细胞中出现明显的变化 ,其中 6个蛋白在肝癌细胞中高表达 ,9个蛋白低表达 ,L0 2细胞中去磷酸化蛋白 17175Da表达较多 ,而在SMMC772 1细胞中磷酸化蛋白172 5 5Da表达较多。结论 SMMC772 1细胞与正常肝细胞L0 2 4蛋白表达差异对指导肝癌诊断和治疗具有重要意义。研究蛋白质的磷酸化 /去磷酸化有助于了解肝癌发生过程和分子机制。 Objective To compare the expression characteristics of proteins in hepatocellular carcinoma cell line SMMC7721 and normal hepatocellular cell line L02 and to screen the distinct proteins for hepatocellular carcinoma.Methods Surface enhanced laser desorption/ionization (SELDI) mass spectrometry with ProteinChip IMAC3 and WCX2 was performed to compare the expressed proteins in SMMC7721 and L02.Protein profiling was read by PBSII C ProteinChip Reader and the proteome database was analyzed by Proteinchip Software 3.0.2.Results Fifteen distinct proteins were found in SMMC7721 cell line in which 6 proteins showed overexpression and 9 proteins underexpression as compared with L02 cell line.The dephosphorylated protein 17175Da expressed higher in L02 cells,but the phosphorylated protein 17255Da expressed higher in SMMC7721cells.Conclusion The overexpressed and underexpressed proteins in protein profiling could be candidate biomarkers of hepatocellular carcinoma.The protein 17175Da phosphorylated/dephosphorylated may be related with the occurrence of hepatocellular carcinoma.
