摘要
目的 建立一种快速、有效的大鼠异位心脏移植模型。方法 实验组 (DA Lewis)和对照组 (Lewis Lewis)各行 2 0例心脏移植。切取大鼠供心时 ,留取主动脉和右肺动脉备吻合 ,而将其余血管分别结扎 ;植入时 ,将供心主动脉和右肺动脉分别与受者的腹主动脉和下腔静脉端侧吻合。术后受体和移植心均存活≥ 5d视为手术成功 ;术后第 6天两组各取 10例行组织学检查。结果 仅对照组 1例失败 ,手术成功率由传统术式的 85 %提高到 97.5 % ,供心缺血时间缩短至 2 0~3 0min。实验组呈急性排斥反应改变。结论 用新法建立模型较传统术式更为快速有效且能明显提高手术成功率。
Objective To establish a quick and effective heterotopic heart transplantation model in rat.Methods There were 20 rats in both experiment group (DA Lewis) and control group (Lewis Lewis) subject to heterotopic heart transplantation.During the procurement of the donor heart,the ascending aorta and the right pulmonary artery were reserved and then anastomosed respectively in the engraftment to infrarenal abdominal aorta and inferior vena cava of the recipient.This method was different from traditional one that used gross pulmonary artery for the vascular anastomosis.The operation was considered be successful if the recipient rat and graft survived over 5 days.Ten animals in each group were randomly chosen for histological examination on day 6.Results Only one rat died in control group after operation.The successful rate of the operation was improved from 85% around of traditional method to 97.5% (39/40).In addition,the ischemic time of donor heart was reduced to 20 30 min.Histologically confirmed acute rejection was found in experiment group.Conclusion The development of model by using our new technique skill is quicker and more effective than the traditional one and results in the successful rate improvement.
