摘要
目的:观察抗痢疾杆菌膜蛋白IpaB和LPS的单抗对痢疾杆菌入侵HeLa细胞细胞骨架的影响。方法:采用不同膜抗原的单抗处理痢疾杆菌后进行HeLa细胞入侵及阻断实验,观察不同膜抗原单抗对痢疾杆菌入侵的作用。结果:观察到抗菌膜蛋白IpaB的单抗和LPS的单抗均不能阻断细菌的入侵且能促进细菌的入侵,单抗作用后受感染的HeLa细胞胞内肌动蛋白极性分布现象更为显著,入侵的细菌数量明显增多。结论:提示痢疾杆菌的膜蛋白和LPS单抗所识别的表位,均具有使细胞骨架发生改变、调节S.flexneri2a 12菌进入HeLa细胞的能力。
Objective:To study the effect of McAbs against S.flexneri2a12 membrane protein and LPS on the bacteria's invasion to the cytoskeleton of Hela cell. Methods:After S.flexneri2a12 treated with McAbs against various membrane antigens, S.flexneri2a12 invaded the HeLa cells. The invasion results were analyzed by laser scanning confocal microscopy. Results:Neither McAbs against S.flexneri2a12 membrane protein IpaB or LPS could not prevent S.flexneri2a12 invasion. On the contrary,both of them could promote the invasion process. In fact, after treatment with these McAbs, actin's polar distribution in infected HeLa cells became more obvious. Conclusion:It suggests that McAbs against membrane protein and LPS recognize such epitopes that are able to regulate cytoskeleton to facilitate the entrance of S.flexneri2a12 into Hela cells.
