摘要
目的 探讨外周血单核细胞 (PBMC)中DcR3基因表达水平与特发性肺间质纤维化 (IPF)的关系。方法 1 4名IPF病人和 1 0名健康对照各取 5ml静脉血 ,肝素抗凝 ,密度梯度离心法分离PBMC ,异硫氰酸胍 酚 /氯仿法提取PBMC总RNA ,用半定量RT PCR法检测PBMC中DcR3基因的相对表达水平 ,并用t检验进行比较。结果 IPF病人组PBMC中DcR3基因的相对表达水平为 0 375± 0 0 88,与正常对照组 (0 2 59±0 0 84)相比有明显差异 (P <0 0 5)。结论 IPF病人PBMC中DcR3基因的表达水平增高。
Objective To investigate the level of DcR 3 gene expression in peripheral blood mononuclear cells (PBMC) and its relation to idiopathic pulmonary fibrosis (IPF).Methods PBMC was isolated from 5 ml heparinized blood from 14 IPF patients and 10 HV (healthy volunteer) by the Ficoll Hypaque density centrifugation method, and the total cellular RNA was extracted using Guanidine Thiocyanate Phenol/Chloroform. RT PCR was used to determine the relative expression of DcR 3 gene in PBMC. Results The relative expression level of the DcR 3 gene in the patients with IPF was higher significantly than that in HV (0.375±0.088 vs. 0.259±0.084, P <0.05 ). Conclusions DcR 3 gene of PBMC over express in patients with IPF.
