摘要
目的 探讨代谢综合征小鼠肝脏改变及肝脏清道夫受体B1 (SR B1 )表达。方法 正常饮食C57BL/ 6J小鼠 1 0只 (对照组 ) ;进高脂高糖食 8w 5只 (实验组 )和 1 6w 1 0只 (实验组 )。测定血脂、血清胰岛素 (INS)及空腹血糖、肝功、肝脏重量、腹部瘦肉及瘦肉含量 ,肝脏脂质含量及SR B1蛋白表达。并做肝脏病理检查。结果 ①实验B组小鼠腹部脂肪量显著高于对照组 (P <0 0 1 )。②实验B组的小鼠血清甘油三酯 (TG)、空腹血糖 (FBG)明显高于对照组 (P <0 0 5) ,胰岛素高于对照组 ,但无显著性 (P >0 0 5)。③实验组小鼠血清TP、ALB及A/G低于对照组 ,其中实验B组小鼠ALB及A/G有显著差异 (P <0 0 5)。血清ALT、ALP及AST高于对照组 ,其中实验B组小鼠ALP有显著差异 (P <0 0 5)。④实验组小鼠肝脏有明显脂肪变性 ,B组重于A组。实验组小鼠肝脏的TC和TG含量显著高于对照组 ,实验B组小鼠达到正常的 2倍以上。⑤实验组小鼠肝脏SB B1表达高于对照组 ,B组强于A组。结论 高脂高糖食C57BL/ 6J小鼠可作为代谢综合征动物模型 ,其存在脂肪肝 ,肝脏的SR B1蛋白表达量高于正常饮食小鼠 ,可能其参与脂肪肝的形成。
Objective To investigate the change of liver and the expression of SR-B1 in the liver of metabolic syndrome mice.Methods Ten normal diet C57BL/6J mice (control group), fifteen high fat and sugar diet female C57BL/6J mice-metabolic syndrome mice model, five fed for 8 weeks (experimental group-A subgroup) and ten fed 16 weeks (experimental group-B subgroup), were measured abdominal fat mass, total cholesterol (TC), triglycerides (TG), high density lipoprotein cholesterol (HDL-C) and fasting blood glucose (FBG) insulin (INS) and liver function, liver weight, content of lipid and the expression of SR-B1 in the livers, pathological change of liver. Results ① The abdominal fat mass of A subgroup was higher than that of normal group ( P <0.01). ② In B subgroup, serum TC and FBG were significantly higher than those of control group ( P <0.05), INS was higher too, but it wasn’t statistically significant ( P >0.05). ③ Mice of experiment group had lower levels of serum TP, ALB and A/G, compared with normal animal, and the difference of serum ALB and A/G ALT of B subgroup was significant ( P <0.05). The levels of serum ALP and AST in experiment group were higher than those of control group, and the difference of serum ALP in B subgroup had statistical significance ( P <0.05). ④ Mice of experimental group had fatty liver; B subgroup was more severe than A subgroup. The content of TG and TC in the liver of experiment group was higher than those of control group. And it reached to 2 times in B subgroup. ⑤ The expression of SR-B1 protein in the liver of experiment group was higher than that of normal mice; B subgroup was higher than A subgroup. Conclusions High fat and sugar diet C57BL/6J mouse had fatty liver and could be a kind of metabolic syndrome mice model. The expression of SR-B1 protein in the liver, perhaps contributed to the fatty liver, was higher than that of normal mice,
