紫杉醇抑制胚胎型横纹肌肉瘤RD细胞株生长的实验研究 被引量:1

摘要 目的探讨紫杉醇对胚胎型横纹肌肉瘤RD细胞株的抗癌作用及其与诱导凋亡的关系。方法将不同浓度的紫杉醇作用于RD细胞,用四甲基偶氮唑蓝(MTT)快速比色法检测其作用的时间效应及剂量效应;在光镜和电镜下观察细胞的形态学变化;琼脂糖凝胶电泳检测DNA片段、用流式细胞仪分析细胞G2?M期分布的改变及凋亡率。结果紫杉醇对体外培养的RD细胞有明显的抑制作用,在一定剂量和时间范围内,其抑制作用呈时间依赖性和浓度依赖性,其24h的半数抑制浓度为(167.4±5.4)ng?ml。100ng?ml紫杉醇作用于RD细胞24h后,透射电镜和DNA琼脂糖凝胶电泳可见到典型细胞凋亡特征。流式细胞仪分析显示紫杉醇可使肿瘤细胞出现G2?M期阻滞并诱导细胞凋亡。结论紫杉醇对横纹肌肉瘤RD细胞有明显的生长抑制作用,并且能诱导RD细胞凋亡,可用于横纹肌肉瘤的治疗。 Objective To observe the anticancer effect of paclitaxel on human embryonal rhabdomyosarcoma cell line RD and its mechanism. Methods RD cells were treated with various concentrations of paclitaxel. Cell morphology, MTT assay, agarose gel electrophoresis, flow cytometry and transmission electron microscopy were performed on paclitaxel-treated RD cells. Results Paclitaxel had a substantial growth inhibitory effect on RD cells. Within a certain range of time and dose, the inhibitory effect displayed a time- and dose- dependent manner. The IC50 of paclitaxel to RD cells for 24 hours was(167.4±5.4)ng/ml. After RD cells were treated with paclitaxel at 100ng/ml for 24h, the typical features of apoptosis were detected by transmission electronic microscopy and agarose gel electrophoresis. The flow cytometry showed G2 / M phase arrest and apoptosis on paclitaxel treated cells. Conclusion Paclitaxel presents a growth inhibitory and apoptosis-inducting effect on RD cells. It might provide a theoretical basis for clinical treatment.
