高效液相色谱法测定清热灵颗粒中黄芩苷含量

Determination of the Content of Baicalin in Qingreling Granules with HPLC

目的 应用高效液相色谱法测定清热灵颗粒中黄芩苷的含量。方法 固定相采用Alltech :AlltimaC1 8色谱柱 (4 6mm× 15 0mm ,5 μm) ;流动相 :甲醇 水 冰醋酸 (5 0∶5 0∶1) ;检测波长 :2 74nm ;流速 :1 0mL·min 1 。结果 黄芩苷在 0 10 3 6～ 0 82 88μg范围内进样量与峰面积呈良好的线性关系 (r =0 9995 ) ,平均加样回收率 99.3 9% ,RSD =1 44 % (n =5 )。结论 该方法测定结果准确、灵敏、重现性好。

Objective To develop an HPLC method for the determination of the content of baicalin in qingreling granules. Methods An Alltech:Alltima C 18 column served as t he chromatographic column. The mobile phase consisted of methanol-water- glaci al acetic acid (50∶50∶1), the flow rate was 1.0 mL·min -1 .The UV detection wavelength was 274 nm. Results There was a good linear relationship between th e concentration of baicalin and absorption area value when the concentrations of baicalin was within the range of 0.103 6-0.828 8 μg (r=0.999 5).The aver age recovery rate was 99.39%, RSD=1.44%(n=5). Conclusion The method was shown to be accurate, sensiti ve, well reproducible and may thus be used in the determination of baicalin in qingreling granules.
