摘要
目的 :探讨脑泰方提取物预处理减轻沙土鼠脑缺血再灌注损伤的作用。方法 :采用钳夹双侧颈总动脉建立沙土鼠脑缺血模型 ,将 6 5只沙土鼠随机分为假手术组 ,脑缺血再灌注损伤组 (IR组 )、脑缺血预处理组 (IPC组 )、脑泰方提取物预处理组 (NPC组 ) ,根据再灌注不同时间 (2 4、4 8、72h和 7天 )。将IR组、IPC组、NPC组各分 4个亚组 (n =5 )。观察各时相沙鼠海马CA1区神经元状况 ,海马CA1区存活神经元数目。结果 :(1)IPC组、NPC组与IR组同期比较 ,海马CA1区存活神经元数目明显增多 (P <0 .0 5 ) ,脑组织结构明显改善 ;(2 )各组不同时相比较 :随着再灌注时间延长 ,海马CA1区存活神经元数目逐渐减少 ,至第 7天达最低 ;(3)药物预处理组与缺血预处理组比较 :海马CA1区存活神经元数目差异无显著性意义 (P >0 .0 5 )。结论
Objective: To investigate the preconditioning effects of Naotai Formula Extract(NTE) on the cerebral ischemic-reperfusion(CIR) injury in gerbils.Methods: CIR model was prepared by bilateral carotid occlution with incarceration method in gerbils. Then, 130 animals were randomly divided into the sham operation group(SOG), cerebral ischemic-reperfusion injury group(IR), cerebral ischemic-preconditioning group (IPC), and NTE preconditioning group (NPC). Furthermore, IRG, IPG and NPC were divided into 4 sub-groups respectively based on the reperfusion duration(24h,48h,72h and 7d), the state of neuron and the number of survival neuron in the hippocampal CA 1 region were observed at different time. Results: (1)Compared with IR at the same time-shape of reperfusion, the number of survival neuron of hippocampal CA 1 region was increased significantly in IPC and NPC group(P<0.05), and its cerebral tissue structure was also improved. (2) Compared various groups at different time, with the Ischemic-Reperfusion time prolonging, the number of survival neuron became low slowly and arrived the lowest at the 7th day in the hippocampal CA 1 region. (3)Compared NPC with IPC group, the number of survival neuron had no distinct significance in the hippocampal CA 1 region (P>0.05). Conclusion: Naotai formula extract preconditioning inhibits the cerebral ischemic-reperfusions injury in gerbil.
