摘要
目的 建立RP-HPLC测定天麻中天麻素含量的分析方法。方法 优化药材的提取及色谱的分析方法得分离良好的色谱图,外标法定量。色谱柱:Hypersil ODS色谱柱(4.6×250mm);流动相:甲醇-0.1%冰醋酸水溶液(5:95);检测波长:270nm。结果 色谱图中12个峰均可用其进行准确定量,其中天麻素含量测定的线性范围为0.60~2.41μg(r=0.9999),平均加样回收率为97.85%,RSD=1.17%(n=5)。结论 本方法简便,准确,重现性好,可用于天麻中天麻素的含量测定,同时也可用于其它水溶性成分的定量。
Objective To constitute the analytical method for determination gastrodin in Gastrodia elata Bl. by RP-HPLC. Method Optimize the extracting and separate method to get the superior spectrum for quantilizing gastrodin by standard curve. HPLC analysis was carried out with C18 column and methanol-0. l%acetic acid (5 : 95) as the mobile phase. The detection wavelength was 270 nm. Result Twelve peaks can be used for quantity, in which gastrodin was determinated with 0. 60-2. 41 μg (r=0. 9999) as the linear range and 97. 85% (RSD=1. 17%, n = 5) as the average recovery. Conclusion The method is simple, reliable and good recurrence. It can be used to determine gastrodin in Gastrodia elata Bl. , and other water soluble constituents as well.
