摘要
目的 :探讨EB病毒 (EBV)转化B淋巴细胞的最佳方法 ,为进一步研究肾综合征出血热 (HFRS)患者的细胞免疫功能奠定基础。 方法 :采用环孢素A(CsA)法、微量全血法和冻存全血法三种方法建立B淋巴母细胞样细胞系 (B LCL) ,比较建立细胞系成功率和所用时间。 结果 :EBV成功转化后的B LCL ,其体积增大并永生化 ,进而增生的淋巴细胞积聚成团。CsA法、微量全血法和冻存全血法转化的成功率分别为 73.3%、2 6 .7%和 4 6 .7%。 结论 :CsA法成功率高 ,建立细胞系所用时间短 ,是建立B LCL的最佳选择。
Objectives:To investigate the optimal method to transform human B lymphocytes(B-LCs)by EB virus(EBV)and lay the foundations for the further study of cellular immunology of patients with hemorrhagic fever with renal syndrome(HFRS). Methods:Using three methods, namely, cyclosporin A (CsA), small amount of fresh whole blood and, freezing whole blood, B-lymphoblastoid cell lines (B-LCLs) were established and their successful rates and time of establishing cell lines were compared among the three methods. Results:The results showed that cellular volume was enlarged and immortalized, followed by the gathering together of multiplied lymphocytes after B-LCs were transformed by EBV successfully. The successful rates of transformation were 73.3%, 26.7% and 46.7%, respectively, in the methods of CsA, small amount of fresh whole blood and freezing whole blood groups. Conclusions:It suggested that the successful rate of CsA method was higher than that of other two methods. The time of establishing cell lines was shorter than that of other two methods. The CsA method was an optimal method to transforme human B-LCs by EB virus. In addition, the factors affecting the successful transformation were discussed in this paper.
