摘要
本文用Lipofectin Reagent作为转染细胞的间型脂质,使Liposome与SV40 ts A209 DNA形成一种混合物转染猪子宫上皮细胞。此法比用磷酸钙和二乙氨乙基葡聚糖转染细胞的方法简单,高效、重复性好。转染后的猪子宫上皮细胞形状和大小略有改变;对温度敏感,用相同培养液培养7天,在33℃条件下细胞的生长速度是在40℃时的5倍.
A method of using lipofectin reagent as a mediated-lipid to transfect pig uterus epithelial cells was described here. The transfector is composed of the lipofectin reagent and ts A209 DNA.
The result showed that this method was simple, highly reproducible and more effective than either the calcium phosphate or DEAE-dextran transfection techniques. The transformed uterus epithelial cells of pig were changed a little bit in shape and size, and were senstive to temperature. The growth rate of the transformed cells at 33℃ was five times faster than that at 40℃ in the same medium for seven-day culture.
