摘要
CICC2 4 6 2是我国酶制剂企业普遍采用的生产菌种 ,由于其不产孢子的特点 ,对该菌种的鉴定目前尚无定论。文中利用分子生物学技术对该菌种进行鉴定 :经菌丝体DNA提取、PCR扩增和序列测定 ,得到ITS1 5 8S ITS2序列 (约 6 0 0bp)。利用BIOEDIT ,TREEVIEW ,CLUSTALX等序列分析软件对该序列进行处理和分析 ,并将序列提交NCBI(NationalCenterforBiotechnologyInformation) ,进行核酸序列同源性比较。经分析认为CICC2 4 6 2为黑曲霉变种 (Aspergillusnigervar )。
CICC2462 is glucoamylase producer which being used commonly by enzyme preparation corporations. As yet, however, the denomination of this strain has not been provided for its unique character of non-spore. In this paper, we adopt molecular biotechnology to certificate the culture.The sequence(600bp) ITS1-5.8S-ITS2 is gotten through the extraction of mycelial, PCR and the examination of sequence. The paper has also used the sequence anaysis software such as BIOEDIT,TREEVIEW and CLUSTAL X to process the sequence, then we submit it to NCBI for homology comparison. At last, the strain was named as Aspergillus niger var.
