摘要
目的 研究腺病毒介导的mIL - 12基因 (AdvmIL - 12 )对裸鼠肝癌生长的抑制作用。方法 用Ad vmIL - 12转染小鼠肝癌H2 2细胞作为实验组 ,以腺病毒转染的H2 2细胞和未转染的H2 2细胞为对照组。将上述细胞分别接种到裸鼠右侧前肢皮下 ,第 11天断头处死裸鼠 ,采血 ,取出瘤组织称重。用ELISA法检测体外细胞培养上清液、接种前后裸鼠血清、肝癌组织中IL - 12表达量。结果 实验组肿瘤平均重量为 (0 87± 0 16 )g ,空白对照组和载体对照组分别为 (1 6 5± 0 39)g和 (1 88± 0 14 )g ,实验组的肝癌平均重量小于对照组 (P <0 0 5 )。AdvmIL- 12 /H2 2细胞上清液中mIL - 12的含量为 (89 71± 2 2 0 5 )ng/ml,Adv/H2 2 ,H2 2细胞上清液中未检测到mIL - 12。接种前各组裸鼠血清中均未检测到mIL - 12的表达。接种后实验组裸鼠血清中仅有少量mIL - 12表达 ,对照组裸鼠血清中未检测到mIL - 12的表达。实验组肝癌组织中mIL - 12的表达量高于对照组 (P <0 0 1) ,对照组之间的mIL - 12平均表达量差异无显著性 (P >0 0 5 )。结论 转染AdvmIL - 12的小鼠肝癌H2 2细胞在裸鼠肝癌组织局部高表达mIL - 12 ,并能抑制裸鼠肝癌的生长。
Objective To study the inhibition of the growth of hepatocarcinoma in nude mice by mIL-12 gene mediated by adenovirus(AdvmIL-12). Methods AdvmIL-12 was transfected into H22 cells of mouse hepatocarcinoma in the experimental group. In the control group, H22 cells were transfected with adenovirus or not transfected. Cells were inoculated into the right forefoot of nude mice. The nude mice were killed on the eleventh day. Blood was collected and the tumor was taken and weighted. The content of mIL-12 in the supernatant of cell culture, nude serum before and after inoculation was measured by ELISA. Results The average weight of tumor was(0.87±0.16)g in the experimental group, and was (1.65±0.39)g and(1.88±0.14)g in the adenovirus control group and blank control group respectively. The weight of tumor in the experimental group was less than that in the control groups(P<0.05). The content of mIL-12 in the supernatant of AdvmIL-12/H22 cells was (89.71±22.05) ng/ml , but mIL-12 was not detected in the supernatant of Adv/H22 cells and H22 cells. mIL-12 was not detected in the serum of any group before inoculation. After inoculation, mIL-12 had little expression in the contral group, but the expression of mIL-12 was not detected in the contral group. The content of mIL-12 of hepatocarcinoma tissue in the experimental group was higher than that of contol groups(P<0.01), but there were no difference(P>0.05) in the control group. Conclusion AdvmIL-12 can highly express mIL-12 in the location of nude hepatocarcinoma in the experimental group and inhibit the growth of nude hepatocarcinoma.
