摘要
为了克服猪肺肝素分离的传统方法操作繁锁、生产周期长、生产成本高、产品品位低的缺点.用高温短时酶解代替低温长时酶解,并对匀浆的最适程度和自溶的最适条件进行了研究。肺组织经匀浆、自溶、酶解、盐解、ND除杂、CPC浓缩、酒精沉淀、丙酮干燥、烘干制得粗品肝素。用该工艺可制得5—6万单位粗品肝素/猪肺,其效价为25.68±1.75u/mg,得率为0.2545±0.0150%。
The new method is studied in order to surmount the shortcomings of the complex operation, the long processing time, the high cost and the low grade. In the new method, the lukewarm temperatured and long timed enzyme digestion,and the optimum homogenization degree and optimum autolysis conditions are found for the first time, the new diminishing impurities method (ND METHOD) is accidentally devised.The crude heparin is made from hoglung, this method includes homogenating, autolysising, digesting by enzyme, digesting by sodium chloride, diminishing impurities by ND, conncentrating heparin by CPG precipitating by alcohol, dehydrating by acetone and drying. 50— 60 thousands IU crude heparin is made from one hoglung by the new method, specific activity being 25.68±.75u/mg and yield 0.2545+0.0150%.
