摘要 目的:探讨环孢霉素A(cyclosporinA,CsA)对角膜碱烧伤房水中炎性细胞因子的影响及作用机制。 方法:将健康家兔32只随机分为4组,其中3组建立Ⅱ度角膜碱烧伤模型,于建模后第1天开始每日分别予球结膜 下注射CsA、地塞米松和生理盐水,另1组为正常组对照。2周后检测房水中肿瘤坏死因子 α(TNF α)和白细胞介 素 1β(IL 1β)含量,并对角膜组织作病理检查,观察炎症反应程度。结果:CsA组房水中TNF α和IL 1β显著低于 正常组(P<0.01),HE染色显示CsA组及地塞米松组炎症反应明显轻于生理盐水组。结论:CsA可减少角膜碱烧 伤房水中TNF α和IL 1β表达及组织中炎性细胞的浸润,提示CsA可用于临床治疗角膜碱烧伤。 Objective: To investigate the effect of cyclosporin A (CsA) on inflammatory cytokine in aqueous of rabbits with cornea alkali burn and its mechanism. Methods: Thirty-two healthy rabbits were randomly divided into 4 groups (8 in each group): control group without alkali burn and 3 experimental groups with models of 20 cornea alkali burn receiving bulbar conjunctiva infection of CsA, dexamethasone and normal saline respectively next day after model establishment. Two weeks later, contents of TNF-α and IL-1β in aqueous humor were determined and the corneal tissue was examined pathologically for the degree of inflammatory reaction. Results: Contents of TNF-α and IL-1β in aqueous humor in CsA group were significantly lower than those in control group (P<0.01). HE staining revealed the inflammatory reaction in CsA group and dexamethasone group was obviously milder than that in normal saline group. Conclusion: CsA may reduce the expression of TNF-α and IL-1β in the aqueous humor of the rabbits with cornea alkali burn and infiltration of inflammatory cells, which indicated the probability of CsA used to treat cornea alkali burn.
