摘要
目的观察极低密度脂蛋白对大鼠系膜细胞分泌FN以及对转化生长因子-β1(TGF-β1)基因转录的作用,进而探讨VLDL对系膜细胞的毒性作用.方法应用ELISA法测定培养细胞上清纤维连接蛋白(Fibronectin,FN)的分泌;RT-PCR法测定系膜细胞TGF-β1的基因表达情况.结果在一定的时间范围内(6~24 h),VLDL可以以时间、剂量依赖性的关系促进系膜细胞分泌FN.随着刺激时间的增加,TGF-β1mRNA转录逐渐增强;VLDL浓度在0~500μg/ml之间时,TGF-β1mRNA转录随着VLDL浓度的增加而逐渐增强.结论VLDL可以促进系膜细胞分泌FN以及TGF-β1的基因转录,提示VLDL具有肾脏毒性.
Objective To observe the effect of very low density lipoprotein on secretion of fibronectin and transforming growth factor-beta 1 gene transcription of rat mesangial cells and to study if VLDL has nephrotoxicity on mesangial cells.Methods ELISA method were employed to determine protein production of fibronectin.RT-PCR method was adopted to determine mRNA transcription of TGF-beta 1.Results Within range of concentration(0~500 μg/ml),VLDL could promote fibronectin production with time and dose dependent manners.Within rather wide range of concentration(10~500 μg/ml),mRNA transcription of TGF-beta 1 were stimulated with time and dose dependent manners.Conclusion VLDL would promote fibronectin production and mRNA transcription of TGF-beta 1 of rat mesangial cells.VLDL may has nephrotoxity.
