摘要
目的 研究肿瘤坏死因子相关的凋亡诱导配体 (TRAIL) 联合三氧化二砷 (As2O3) 诱导急性早幼粒白血病细胞株HL 60凋亡, 探讨其应用于临床的可行性。方法 在对数生长期的 HL 60 细胞中分别加入不同浓度的TRAIL和1μmol/L的As2O3, 采用MTT比色法测定TRAIL和 As2O3 单独和联合应用时对细胞的生长抑制率, 并用流式细胞术检测细胞凋亡。结果 TRAIL联合 As2O3 可协同降低 HL 60 细胞活力, 抑制细胞增殖, 诱导细胞凋亡。当TRAIL浓度小于100μg/L时, 其作用随培养时间延长及药物浓度增加而增强 (P<0 .05), 而当 TRAIL浓度再增高时, 对细胞生长抑制率和凋亡率的增强作用不明显 (P>0 .05)。结论 TRAIL与 As2O3 具有协同作用, 能高效杀灭白血病细胞。TRAIL是一种有望应用于白血病临床治疗的新型生物制剂。
Objective To investigate the apoptosis-inducing effect of TRAIL and arsenic trioxide (As 2O 3) on leukemic cell line HL-60,and study the feasibility of TRAIL to be used in clinical practice.Methods Various concentrations of TRAIL and 1 μmol/L As 2O 3 were added in exponential growth HL-60 cells respectively.MTT assays were used to measure the growth inhibitory effect of combined use of TRAIL and As 2O 3 or alone.Flow cytometry was used to evaluate apoptosis.Results Combined use of TRAIL and As 2O 3 could synergistically inactivate HL-60 cells,inhibit the cell proliferation and induce apoptosis in a dose- and time-dependent manner when the concentration of TRAIL was less than 100 μg/L (P<0.05).The increase of growth inhibitory rate and apoptosis rate were not significant when its concentration was over 100 μg/L (P>0.05).Conclusion TRAIL can present a synergistic effect with As 2O 3.TRAIL has a promising prospect in clinical use of leukemia.
