三宝胶囊对老年大鼠抗氧化能力的影响

Effect of sanbao capsule on antioxidant ability of old rats

目的:通过三宝胶囊对老年大鼠抗氧化能力的影响,探讨三宝胶囊的抗衰老作用。方法:实验于2003-04/07在山西师范大学体育学院动物实验室、山西师范大学生命科学学院实验室、山西省地方病研究所实验室完成。青年雄性SD大鼠(青年对照组)8只,老年雄性SD大鼠16只,随机分为老年对照组8只,老年中药组8只。老年中药组灌服三宝胶囊,按4粒/kg剂量给药,溶于2.0～3.0mL自来水中。青年对照组、老年对照组灌服同体积的自来水。第9周末取样测定血清、心肌、肾脏中丙二醛和超氧化物歧化酶(superoxidedimutase,SOD)、谷胱甘肽过氧化物酶(glutathioneperoxidase,GSH-Px)活性以及谷胱甘肽含量。结果:老年对照组与青年对照组相比,血清、心肌组织SOD酶活性减低(P<0.01),心肌组织、肾组织GSH-Px酶活性减低(P<0.05)。老年中药组与老年对照组比较,血清SOD酶活性升高(P<0.01),心肌组织SOD酶活性有升高的趋势,但无统计学意义(P>0.05)。老年对照组与青年对照组相比,血清、心肌组织、肾组织丙二醛含量升高(P<0.05),心肌组织、肾组织谷胱甘肽含量减低(P<0.01)。老年中药组与老年对照组比较,心肌组织、肾组织谷胱甘肽含量升高(P<0.01),血清、心肌组织丙二醛含量有减低趋势,但无统计学意义(P>0.05)。老年中药组与老年对照组比较。

AIM:To investigate the effect of sanbao capsule on antioxidant ability of old rats so as to probe into the antiaging effect of sanbao capsule. METHODS:The experiment was conducted in the laboratory for animals of College of Physical Education,the laboratory of College of Life Sciences in Shanxi Normal University,and the laboratory of Shanxi Institute for Endemic Diseases.Twenty-four SD rats were randomly divided into 3 groups:young control group,old control group and Chinese-medicine-taken old group with 8 rats in each group.Rats of Chinese-medicine-taken old group were perfused with sanbao capsule at the dose of 4 capsules per kilogram dissolving in 2.0 to 3.0 mL tap water.The rats of young and old control group were fed with tap water in the same volume.At the end of the ninth week,all rats were sacrificed to determinate the activities of malondialdehyde(MDA),superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in serum,kidney and cardiac muscle and the concentration of glutathione(GSH). RESULTS:Compared with those in the old control group,the activities of SOD in serum and cardiac muscle were significantly decreased(P< 0.01),and the activities of GSH-px in cardiac muscle and kidney were significantly decreased(P< 0.05) in the young control group.In the old control group,the activity of SOD in serum was significantly higher(P< 0.01),and that in cardiac muscle had a increase tendency,but without any significant difference(P >0.05).The concentrations of MDA in serum,cardiac muscle and kidney were increased in the old control group as compared with those in the young control group(P< 0.05),and the concentrations of GSH in cardiac muscle and kidney were decreased in the former group,as compared with those in the latter group(P< 0.01).The concentrations of GSH in cardiac muscle and kidney in the Chinese-medicine-taken group were decreased as compared with those in the old control group(P< 0.05),and the concentrations of MDA in serum and cardiac muscle had a decreasing tendency in the former group,but without any significance(P >0.05). CONCLUSION:Sanbao capsule is effective in increasing the activity of antioxidant enzymes and decreasing lipid peroxidation.
