摘要
目的:探讨血管内皮生长因子及分泌型磷脂酶A2对谷氨酸诱导增强的神经元死亡的不同作用。方法:原代胚胎鼠皮质神经元细胞培养,细胞毒性试验及生化指标测定,细胞染色,显微镜下观察形态学改变及影像分析。结果:不同浓度的谷氨酸12.5、25、50、100μM可单独增加胚胎鼠皮质神经元死亡且呈剂量及年龄依赖关系。V亚型分泌型磷脂酶A2可协同增强谷氨酸所致神经元死亡。不同浓度的血管内皮生长因子10、30、100 ng/ml对分泌型磷酯酸酶A2协同增加的谷氨酸神经元毒性有解除作用且呈剂量依赖关系。结论:血管内皮生长因子可解除分泌型磷脂酶A2协同增加的谷氨酸诱导的皮质神经元毒性作用。
Objective: To discuss the different function between the Vascular endothelial growth factor (VEGF) and group v secreted Phospholipase A2 ( sPLA2 ) for Cortical neurons for Glutamate - induced toxicity. Methods: Primary embryonic rat cortical neurons culture, cytotoxicity assays and biochemistry detection, cell stain for the neuron morphology in microscopy and image analysis. Results: Different concentration of glutamate 12.5 , 25, 50, 100μM alone increased the embryonic rat cortical neuronal cell death and in a concentration - dependent manner and all effects are age - dependent relation. sPLA2 increased glutamate - induce neuron. Different concentration of VEGF 10,30, 100 ng/ml quit the Glutamate - induce neuron toxicity potentiated by sPLA2 and in a concentration - dependent manner. Conclusions: VEGF rescues embryonic rat cortical neurons from sPLA2 potentiating glutamate induced neuronal cell death.
