摘要
目的 探讨整合素β3反义寡核苷酸(ASODN)活体转染假孕小鼠子宫内膜抗早孕的可能性。方法 将 整合素β3反义寡核苷酸注入假孕1 d小鼠子宫,同时同侧输卵管植入2细胞期受精卵10枚,小鼠正常饲养,5 d 天取小鼠子宫内膜进行Western Blotting检测整合素β3蛋白表达情况;19 d进行小鼠子代出生率评估。结果 Western blotting检测5 d小鼠子宫内膜整合素β3蛋白强表达,10 pmol整合素β3反义寡核苷酸转染假孕小鼠子 宫内膜后5 d小鼠子宫内膜整合素β3蛋白减弱表达,100 pmol整合素β3反义寡核苷酸转染时弱表达,单纯脂质 体轻度影响其表达;19 d进行小鼠子代出生率评估,受孕鼠转染整合素β3反义寡核苷酸后子代小鼠出生抑制率 为85%。结论 整合素β3反义寡核苷酸可能整合到假孕小鼠子宫内膜抑制其蛋白表达,阻止胚胎着床。由于 例数较少,其整合素β3反义寡核苷酸后抑制子代小鼠出生率需进一步研究。
Purpose To investigate the possibility of resisting early pregnance when the endometrosis of pseudo-pregnancy mice vivo-transfected by integrin-β3 ASODN. Methods Integrin-β3 was infused to the endometriosis of 1 day pseudo-pregnancy mice, at the same time,the fertilized eggs were implanted to the homolateral duct. The mice were bred normally. Five days later, the expression of integrin-β3 albumen in the endometriosis was detected and the natality of filizl generation was estimated after 19 days. Results There was strong expression of integrin-β3 in mice's endometriosis detected by Western blotting. When pseudopregnancy mice transfected by 10 pmol integrin-β3 ASODN, the expression of integrin-β3 in the indometriosis was weakened meanwhile no expression when 100 pmol did. Simple lipin didn't affect the expression. The natality of filial generation was estimated 19 days later. The inhibition rate of filial generation was 85 % when transfected by integrin-β3 ASODN. Conclusions Integrin-β3 ASODN could be integrated to the endometriosis of pseudo-pregnancy mice, and inhibited the expression of its albumen, then prevent the embryo's implantation.
