摘要
目的 :探讨皮肤鳞状细胞癌 (简称鳞癌 )中基质金属蛋白酶 2 (MMP -2 )、金属蛋白酶组织抑制因子 2 (TIMP-2 )的表达及其与肿瘤分化程度和淋巴结转移的关系。方法 :用ABC免疫组化技术观察5 0例皮肤鳞癌 (高分化鳞癌 3 4例 ,低分化鳞癌 1 6例 ,其中 1 4例淋巴结转移 )。结果 :MMP -2与TIMP-2均表达于瘤细胞和癌旁间质细胞、血管内皮细胞质中。MMP -2表达阳性率为 76% ( 3 8/ 5 0 ) ,在低分化鳞癌组中的表达明显高于高分化组 (P =0 0 2 1 )和有淋巴结转移组 (P =0 0 0 3 )。TIMP -2表达阳性率为74% ( 3 7/ 5 0 ) ,在高分化或无淋巴结转移的鳞癌组中的表达明显高于低分化(P =0 0 1 0 )或有淋巴结转移组(P =0 0 1 8)。结论 :皮肤鳞癌中MMP -2和TIMP
Objective:To investigate the expression of matrix metalloproteinase 2 (MMP-2), tissue inhibitor of metalloproteinase 2 (TIMP-2) in cutaneous squamous cell carcinoma(SCC) and the relationship between their expression, tumor differentiation and lymph node metastasis. Methods:The expression of MMP-2,TIMP-2 were observed by ABC imunohistochemiccal techique in 50 cutaneous SCC (including 14 SCCs with lymph node metastases), among them there were 34 highly differentiated and 16 poorly differentiated ones. Results:MMP-2 and TIMP-2 were localized within cytoplasm of tumor cells, stromal, blood vessel and endothelial cells surrounding tumor nests. MMP-2 expressive rate was 76%, expression in poorly differentiated tumors without lymph node metastasis was significantly higher than that in highly differentiated ones(P=0.021) and with lymph node metastasis(P=0.003). TIMP-2 expressive rate was 74%, and the expression in well differentiated SCC or that without lymph node metastasis were significantly higher than those inpoorly differentiated ones(P=0.010) or that with positive metastasis(P=0.018). Conclusion:Expressions of MMP-2 and TIMP-2 were showed the positive relationship between the differentiation and lymph node metastasis in cutaneous SCC.
