摘要
目的 :探讨唾液蔗糖酶活性与口腔肿瘤恶变之间的相互关系。方法 :采用邻甲苯胺测糖法 ,测定单位时间内唾液蔗糖酶水解蔗糖生成的葡萄糖量 ;在PH6 ,37℃条件下进行酶反应用双倒数作图法测出表观Km和表观Vm值 ;检测蔗糖酶活性。结果 :正常人唾液蔗糖酶活性为 1 32 2± 0 434mU/mgpro ,口腔鳞癌患者唾液蔗糖酶活性为 1 5 48± 0 6 5 7mU/mgpro ,二者之间比较无显著差异 (P >0 0 5 ) ;正常人唾液蔗糖酶的表观Km =(1 1 2± 2 4)× 1 0 -2 mol/L ,表观Vm =1 1 38± 0 35 2 (每分钟光密度值 ) ,口腔鳞癌患者唾液蔗糖酶的表观Km =(9 5±2 2 )× 1 0 -2 mol/L ,表观Vm =(1 0 89± 0 6 32 ) (每分钟光密度值 )。结论 :口腔良性肿瘤、涎腺腺瘤、口腔鳞癌患者与正常人比 ,唾液蔗糖酶活性无显著差异。
Objective:To investigate the relation between the activity of saliva-sucrase and malignant change of oral cavity tumor.Methods;The activity of sucrase was detected by way of measuring the quality of glucose producing by hydrolysis of saliva-sucrase to sacrose in a definite period with toluidine.The Km and Vm with a set of double-reciprocal plot were obtained after carrying on enzyme reactions under the condition of pH 6 and 37℃。Results:The activity of saliva-sucrase of normal person was 1.322±0.434mU/mg pro,patient of oral cavity squamous cell carcinoma was 1.548±0.657mU/mg pro and there was no remarkable difference between them (p>005).The Km and Vm of saliva-sucrase of normal person were (11.2±2.4)x10 -2 mol/L and 11.2±2.4(optical density perminute).They were (9.5±2.2)×10 -2 mol/L and 1.089±0.632(optical density perminute) in oral cavity squamous cell carcinoma patient.Conclusion:There was no notable disparing of the activity of saliva-sucrase between them comparing patient of oral cavity squamous cell carcinoma with normal person.So did with the patient of oral cavity tumor and salivary adenoma.
