地龙提取物对实验性血栓的溶解作用及有效成分的研究 被引量:15

摘要 地龙组织水溶性提取物中含有能够溶解家兔实验性血栓和人全血凝块、多血小板血浆凝块及人纯纤维蛋白凝块的“纤溶酶样”物质(称地龙溶栓酶)。体外实验,地龙组织粗提取液的溶栓时间为5.55±0.08小时,蛋白提取液的溶栓时间为5.67±0.12小时。DEAE纤维素及Sephadex G-50柱层析分别显示5个蛋白峰和4个蛋白峰,各峰洗脱液的溶栓实验结果表明溶栓有效成分存在于DEAE纤维素柱层析的第5蛋白峰和Sephadex G-50拄层析的第2蛋白峰。Sephadex G-50柱层析第2蛋白峰洗脱液的聚丙烯酰胺凝胶电泳显示7条区带,其中有3条区带具有明显的溶栓作用。 In the water-soluble extracts from the earthworm tissue there are substances like fibrinolytic enzyme(called thrombolytic enzyme) which can dissolve the experimental thrombus of rabbit, the clots of human blood and purified fibrin. In vitro? the thrombolytic time of the crud extracts of the earthworm tissue is 5.55±0.08 h, protein extract 5.67±0,12 h. The chromatography of the DEAE cellulose column shows five peaks and the fifth peak has thrombolytic effect. The chromatography of the Sephadex G-50 column shows four peaks and the second peak has thrombolytic effect. The.eluant of the second peak of Sephadex G-50 shows seven bands by PAGE, of which three bands have strong thrombolytic action.
