结核杆菌对利福平耐药的分子机理的研究 被引量:1

摘要 目的 :研究本地区临床分离的结核杆菌利福平 (RFP)耐药表型与rpoB基因突变的关系。方法 :应用全自动DNA测序仪对 1株RFP敏感株和 2株RFP耐药株的rpoB基因的突变集中区域进行了测序分析。结果 :RFP敏感株证实无rpoB基因突变 ,而 2株RFP耐药株均发生rpoB基因的碱基置换突变 ,1株为Leu511→Val ,Ser53 1→Leu ;另一株为Ser512 →Gly。结论 Objectve:For the sake of studying the relationship between rpoB gene mutations and rifampin resistance.Method:The hot region in the rpoB gene of 1 RFP susceptible strain and 2 RFP resistant strains were analyzed by automated DNA sequencing.Results:There is no mutation in rpoB gene of 1 RFP susceptible strain,But the distinct substitutions,Leu to Val change in codon 511,Ser to Leu change in condon 531 and Ser to Gly change in codon 512 accounting for 2 RFP resistant isolates,were identified.Conclusion:The substitutions of highly conserved aminoacids encoded by rpoB gene appears to be the molecular mechanism responsible for RFP resistance.
