摘要
几丁酶(EC 3·2·1·14)能将几丁质分解为寡聚N-乙酰胺基葡萄糖或N-乙酰胺基葡萄糖。几丁质是许多真菌细胞壁的主要成分。在高等植物中未发现几丁质的存在,但却广泛存在有几丁酶。近年来的研究表明,植物中的几丁酶在植物对病原真菌侵染所作出的保卫反应中有重要的作用。植物几丁酶的性质与功能已引起人们的兴趣和重视。苦瓜(Momordica charantia L.)是一种病害很少的蔬菜作物,其几丁酶的纯化及性质研究尚未见报道。为了探讨苦瓜抗病性的蛋白质基础,本文对此进行了研究。
Chitinase was purified from Momordica charantia L. by affinity chromatography. The purified enzyme showed single band on sodium dodecyl sulfate polyacrylamide gel electrophoresis and the molecular weight was estimated as 35 kD. The enzyme was stable at temperatures up to 50℃ or less than 10% loss of actuvity in l h. Its optimum temperature was about 45℃.Its suitable pH had a rather wide range from pH 4.4 to pH 6.8 and the optimum pH was about 6.2. The activity of the enzyme was similar in root and stem. In the lower leaves,the activity was higher than that of the upper.
