摘要
利用N-溴代琥珀酰亚胺化学修饰叶绿体腺三磷酶,测得该该酶含3.5个色氨酸残基,其中2个色氨酸残基与酶活性有关。光氧化腺三磷酶伴随吸氧量增加,酶活性逐渐降低,经NBS修饰或光氧化的腺三磷酶,其免疫抗原减弱。表明腺三磷酶中色氨酸基参与酶的活性和抗原性。
The numbers of tryptophane residues in spinach CF_1-ATPase were measured by means of chemical modification with N-bromosuccinimide (NBS) and photooxidation. There are 3. 5 tryptophane residues in CF_1-ATPase, among which two are essential for the enzyme activity. Photooxidation of CF_1-ATPase led to increased O_2 uptake of the reaction system and loss in activity of CF_1-ATPase . Immunological property of CF_1-ATPase has been altered by chemical modification with NBS and photooxidation. The results show that tryptophane residues seen to be essential for activity and antigenic properties of CF_1-ATPase .
