TGF-β对原代培养肝细胞及肝癌细胞BEL7401生长作用的影响 被引量:1

摘要 目的 研究转化生长因子-β(TGF-β)对正常大鼠肝细胞和肝肿瘤细胞的影响。方法 分离和培养原代大鼠肝细胞,培养BEL401肝癌细胞,通过测定细胞DNA合成、细胞活性(MTT)及碱性磷酸酶活性,研究TGF-β对正常大鼠肝细胞和肝肿瘤细胞的影响方式及可能的作用途径。结果 实验发现,TGF-β可以刺激BEL7401肝癌细胞的增生,促进其DNA合成,并增强其活性STGF-β对原代培养肝细胞DNA合成无明显影响,对其活性有明显的抑制作用。实验还发现,TGF-β明显刺激BEL7401肝癌细胞中的碱性磷酸酶增高,同时抑制原代培养肝细胞的碱性磷酸酶活性。结论TGF-β对正常细胞和肝癌细胞BEL7401的生长有不同的影响作用,这种结果可能是通过不同的细胞信号传导途径实现的。 Objective To study effect of transforming growth factor-β (TGF-β) on rat hepatocyte and liver cancer cell BEL7401 Method Primary hepatocyte was prepared and cultured in DMEM culture medium with TGF-β, while the liver cancer cell BEL7401 was cultured at the same condition in different culture pool. Fina1ly, DNA synthetization, cell activity by MTT and alkaline phosphatase activity were assessed to research how TGF-β effects on cells. Result In primary hepatocyte and liver cancer cell BEL7401 culture, TGF-β stimulated BEL7401 DNA's synthesis but had not obvious effect on primary hepatocyte; it also stimulated the cell activity of BEL7401 and suppressed the primary hepatocyte. TGF-β markedly raised the activity of BEL7401 cell's alkaline phosphatase, in the meantime, it suppressed that of primary culture hepatocyte's. Conclusion These data imply that TGF-β has different effects on the normal liver cc11 and BEL7401 cell, this may result from different ways of cell. signal conductions.
